Development and validation of an ELISA for the quantification of bovine ITIH4 in serum and milk.

This paper published in Veterinary Immunology Immunopathology includes the validation of our ELISA for bovine ITIH4.

The aim of this study carried out in collaboration with the University of Zaragoza, was to develop an ELISA for bovine ITIH4 that would allow the determination of this biomarker in both serum and milk. ITIH4 is a new acute phase protein in cattle, described for the first time at the biochemistry department of the University of Zaragoza. It is homologous to Pig-MAP, one of the main biomarkers of inflammation and infection in pigs. In addition to the serum, ITIH4 is present in the milk of cows with mastitis, although in a much lower concentration.The previously available tests did not have the necessary sensitivity to quantify these samples. The ELISA we have developed has adequate sensitivity and precision for both serum and milk measurements (intra- and inter-assay CV less than 10% in both cases). It quantifies proportionally in the working range and shows a good agreement with the reference method (radial immunodiffusion). It is therefore an accurate and precise method valid for serum and milk samples.
